Answer to all three questions: No you can't. The coders are working on it, but it's nothing releasable yet; and Nintendo seems to have caught on and introduced countermeasures, so it'll take some time.

DeSmuME only uses the .dsv file format for saving, which is basically a RAW savefile with a footer containing savetype-related data. If you want a RAW .sav file, either delete the footer using a text/hexadecimal editor, or use the "File->Export Backup Memory" menu item.

For other questions, you should take a look at the FAQ. The stickied [Official Thread] has the link to it.

You can connect with your friend with DeSmuME, just not with Nintendo Games.

DeSmuME to DeSmuME, different computers, via Wi-Fi.
DeSmuME to DeSmuME, same computer, via Wi-Fi.
    DeSmuME to DS, via Wi-Fi.

Basically DeSmuME has the early stages of Wi-Fi support. It can send and receive packets via the internet. It is just that its not 100% emulated and Nintendo can detect something it not right with it and block DeSmuME from the Nintendo Wi-Fi Connection Servers. You can use DeSmuME with homebrew games that don't use Nintendo's Servers just fine.

Do u mean we do that to the .dsv file or can we change the DeSmume internal or ini file like we can for No$cash in the ini file?

to the dsv file....however it's much better to just use the File->Export Backup Memory option...works like a charm.
